Background InformationProtein 33K (UniProt: P24940; also known as L4-33K, Splicing factor 33K, Terminase, small subunit) is encoded by the L4 gene (Gene ID: 2653005) in human Adenovirus 5. Adenovirus 5. Adenovirus 5 belongs to the Adenoviridae family of non-enveloped viruses that are characterized by linear, double-stranded DNA genomes. Their capsid is an icosahedron that contains at least 12 different viral proteins. Adenovirus 5 is commonly used for gene transfer experiments due to its ability to infect a wide group of different cell types. It can harbor large genes in its genome incorporated via homologous recombination techniques. L4-33K is a homooligomeric protein that serves as a virus-encoded alternative RNA splicing factor. It is expressed in the late phase of viral replicative cycle. It activates splicing of viral late gene transcripts that contain weak 3 splice sites. It contains a short arginine-serine repeat region (aa 178-198) that is essential for its splicing enhancer function. L4-33K also plays a role in adenovirus assembly and in adenovirus morphogenesis by promoting the packaging of the viral genome into the empty capsid. It is essential for the temporal activation of major late pre-mRNA splicing at late stages of infection and it induces the splicing and expression of the late capsid vertex protein. It is suggested to function as the small terminase that is part of the molecular motor that translocates genomic DNA in empty capsid during DNA packaging. It is not detected in mature virions. The L4-33K protein may be phosphorylated by DNA-dependent protein kinase (DNA-PK) and protein kinase A (PKA). Its phosphorylation by DNA-PK is reported to have an inhibitory action in L4-33K-mediated alternative RNA splicing whereas the phosphorylation by PKA stimulates L4-33K-mediated alternative RNA splicing.
