Background InformationTargeting protein for Xklp2 (Tpx2) is a major spindle assembly factor belonging to the TPX2 family of proteins. Tpx2 is a required component in the normal assembly of mitotic spindles and, during apoptosis, for microtubules. It is also necessary to the nucleation of microtubules in a chromatin and/or kinetechore dependent manner. Tpx2 activates Aurora-A kinase through promotion of autophosphorylation, protects the activated kinase from dephosphorylation, and regulates its localization to spindle microtubules. Normal expression of Tpx2 is exclusive to proliferating cells, specifically those transitioning from G1/S to the termination of cytokinesis. Tissue expression has only been observed in lung cancer cell lines. Phosphorylation of Tpx2 occurs on serine residues; however, the purpose of the modification has yet to be determined.
