Biological Information
ImmunogenHuman MEK 5 synthetic peptide (aa. 59-74; Genbank Accession # U71088), conjugated to KLH; identical to rat MEK5 sequence over these residues.
ConcentrationPlease refer to the Certificate of Analysis for the lot-specific concentration.
HostRabbit
SpecificityMEK 5 is also known as MAP Kinase Kinase 5, or ERK Kinase 5. AB3184 detects a 49 kDa protein, corresponding to the expeted molecular mass of Mek5, in rat brain, rat liver, rat kidney, rat heart, mouse brain, and mouse testis tissue extracts as well as in cell lysates prepared from the rat pheochromacytoma (PC12) and the human HeLa cell lines. No cross-reactivity with MEK 1, 2, 3, 4 or 6 has been observed with this antibody.

Entrez Gene Number
Entrez Gene SummaryThe protein encoded by this gene is a dual specificity protein kinase that belongs to the MAP kinase kinase family. This kinase specifically interacts with and activates MAPK7/ERK5. This kinase itself can be phosphorylated and activated by MAP3K3/MEKK3, as well as by atypical protein kinase C isoforms (aPKCs). The signal cascade mediated by this kinase is involved in growth factor stimulated cell proliferation and muscle cell differentiation. Four alternatively spliced transcript variants of this gene encoding distinct isoforms have been described.

UniProt Number
UniProt SummaryFUNCTION: SwissProt: Q13163 # May participate together with ERK5 in a growth factor stimulated signaling cascade. Specifically activates ERK5, but not other MAP kinases such as ERK2 or p38. Is not phosphorylated by RAFA, RAFB or RAFC. May interact with GTPases such as CDC42.
SIZE: 448 amino acids; 50112 Da
SUBUNIT: Interacts with PARD6A and MAP3K3. Forms a complex with SQSTM1 and PRKCZ or PRKCI (By similarity). Interacts with Yersinia yopJ.
TISSUE SPECIFICITY: Expressed in many adult tissues. Abundant in heart and skeletal muscle.
DOMAIN:SwissProt: Q13163 The OPR domain mediates interactions with SQSTM1, PARD6A and MAP3K3 (By similarity).
PTM: Activated by phosphorylation on Ser/Thr by MAP kinase kinase kinases (By similarity). & Yersinia yopJ may acetylate Ser/Thr residues, preventing phosphorylation and activation, thus blocking the MAPK signaling pathway.
SIMILARITY: Belongs to the protein kinase superfamily. STE Ser/Thr protein kinase family. MAP kinase kinase subfamily. & Contains 1 OPR domain. & Contains 1 protein kinase domain.
