Background InformationUbiquitin thioesterase otulin (EC:3.4.19.12; UniProt Q96BN8), also known as Deubiquitinating enzyme otulin, Gumby, OTU domain-containing deubiquitinase with linear linkage specificity, Protein FAM105B, Ubiquitin thioesterase Gumby), is encoded by the OTULIN gene in murine species. Gumby belongs to the ovarian tumour (OTU) domain-containing protease family of deubiquitinases (DUBs) that catalyze the removal of ubiquitins from ubiquitinated proteins. Gumby is reported to associate with linear ubiquitin assembly complex (LUBAC) and counteract LUBAC activity by removing linear ('Met-1'-linked) polyubiquitin chains from substrate proteins. The gumby–LUBAC axis plays an important role in angiogenesis, craniofacial and neuronal development by regulating canonical Wnt signalling.
