Key Applications
  • ELISA
  • Western Blotting
  • Immunohistochemistry
Application NotesWestern blotting: 1-10 μg/mL using ECL.

Immunohistochemistry: 5-10 μg/mL It has been reported that the antibody will recognize CART in neurons (cell bodies) from the rat hypothalamic arcuate nucleus.

ELISA: 1:10,000-1:50,000 using 50-100 ng control peptide (Catalog Number AG228) per well.

Optimal working dilutions must be determined by the end user.
Biological Information
ImmunogenMouse/rat CART peptide (55-102).
ConcentrationPlease refer to the Certificate of Analysis for the lot-specific concentration.
HostRabbit
SpecificityRecognizes Cocaine- and Amphetamine Related Transcript (CART). CART has been found to control satiety modulating the actions of two key regulators of food intake, leptin and NPY. Starvation decreases CART levels in the arcuate nucleus (Kristensen et al. 1998). Obese animals have virtually no CART. CART inhibits both normal and starvation-induced feeding, and completely inhibits NPY-induced feeding. Immunoneutralization of CART by administration of anti-CART resulted into higher food intake suggesting that CART is an endogenous regulator of food intake.

Antibody TypePolyclonal Antibody
Entrez Gene Number
Entrez Gene SummaryCART appears to have roles in reward, feeding, and stress, and it has the functional properties of an endogenous psychostimulant (Kuhar et al., 2002 [PubMed 12147208]).[supplied by OMIM]

UniProt Number
UniProt SummaryFUNCTION: SwissProt: Q16568 # Satiety factor closely associated with the actions of leptin and neuropeptide y; this anorectic peptide inhibits both normal and starvation-induced feeding and completely blocks the feeding response induced by neuropeptide Y and regulated by leptin in the hypothalamus. It promotes neuronal development and survival in vitro.
SIZE: 116 amino acids; 12829 Da
SUBCELLULAR LOCATION: Secreted (Potential).
TISSUE SPECIFICITY: Hypothalamus. Found in neurons of the ventrolateral part of the arcuate nucleus, in the external zone of the median eminence, and also found in terminals in the periventricular part of the paraventricular nucleus.
DISEASE:SwissProt: Q16568 # A defect in CART is associated with reduced resting energy expenditure and cosegregates with obesity phenotype.
SIMILARITY: SwissProt: Q16568 ## Belongs to the CART family.
