DescriptionProtein A purified mouse monoclonal antibody. Recognizes poly-ADP-ribose.
BackgroundThis monoclonal antibody is directed against poly(ADP)-ribose (PAR) which is synthesized by the nuclear DNA repair enzyme poly(ADP)-ribose polymerase (PARP). PARP is selectively activated when DNA strands break and catalyze the addition of long branched chains of PAR to a variety of nuclear proteins, such as PARP. The amount of PAR formed in living cells with DNA damage is directly related to the extent of the damage. When DNA is damaged, PAR undergoes a rapid turnover, with a half-life only minutes long, as it is rapidly hydrolyzed and converted to free ADP-ribose by the enzyme poly(ADP)-ribose glycohydrolase (PARG). Upon DNA damage by γ-irradiation or oxidative stress PAR fluorescence can be detected for approximately 10 min. PARP is strongly activated during the so-called execution phase of apoptosis, causing a second round of PAR synthesis. PAR synthesized early in apoptosis is remarkably stable and its immunofluorescence correlates well with other markers of apoptosis.
