A bacterial protein toxin that binds to and enters eukaryotic cells via receptor-mediated endocytosis. Acts intracellularly to initiate the inositol trisphosphate signaling pathway, Ca2+ mobilization, and DNA synthesis. Stimulates tyrosine phosphorylation of multiple substrates including focal adhesion kinase. Exerts its action on the α-subunit of the Gq family of G-proteins. Has been used for the activation of Gq-protein-coupled phosphatidylinositol-specific phospholipase C (PLC) in intact cells, making PMT useful as a tool for studying Gq-linked signal transduction.

      Revision18-August-2008 RFH
DescriptionA dermatonecrotic toxin produced by some strains of Pasteurella multocida. PMT binds to and enters mammalian cells via a receptor-mediated endocytosis mechanism that is not well defined. Causes a transient activation of G-protein, Gqα. Activates phospholipase C and stimulates the formation of inositol phospholipids and diacylglycerol that results in Ca2+ mobilization from an intracellular pool and translocation of protein kinase C to cell membranes. PMT is also reported to stimulate tyrosine phosphorylation of focal adhesion kinase (FAK125) and paxillin. Reported to activate stress fiber formation and increase trans-endothelial permeability by stimulating Rho kinase and myosin light chain phosphatase.
