Background InformationBreast cancer is the most common malignancy in women throughout the world. The global incidence rate has been increasing annually at a rate of 3.1%. Globally, about 2.1 million women were diagnosed with new occurrences of breast cancer and 626,679 women died with breast cancer in 2018. Higher reported incidence rates in wealthier regions may be explained by enhanced access to medical care and diagnostic technologies. Early diagnosis often results in a more positive prognosis.

HER2/neu is an important gene implicated in various cancer types, but particularly in breast cancer. It is a part of the epidermal growth factor receptor family (EGFR). Its protein product is a p185 tyrosine kinase growth factor receptor that is homologous to other EGFRs. When overexpressed or mutated, the extracellular domain will often form homodimers or heterodimers with other EGFRs. Overexpression has typically been correlated with poorer prognosis, higher invasiveness, and can be used to predict the response of certain treatments. HER2/neu gene amplification or overexpression is present in 25-30% of invasive breast carcinomas. The involvement of p185 in tumor progression and initiation has therefore made it a target of interest for therapeutics.

      Source
The TUBO line is a cloned cell line that was established in vitro from a lobular carcinoma that arose spontaneously in a BALB-neuT mouse.

      Applications
ApplicationRat p185 is xenogeneic in BALB/c mice, differing in <6% of the amino residues from mouse p185.1 In the rat-p185 protein, a single point mutation results in the transformation of the Her-2/neu protooncogene to a dominant oncogene. The TUBO cell line has also been shown to express MHC class I glycoproteins, but not MHC class II. The use of the TUBO cell line enables further research into the mechanisms of breast cancer development and potential for new therapies or treatments.
