      Background InformationRegulator of G-protein signaling 14 (UniProt: O08773; also known as RGS14) is encoded by the Rgs14 gene (Gene ID: 114705) in rat. RGS14 is expressed at high levels in the brain cortex, hippocampus, striatum, thalamus and substantia nigra. Its expression has also been reported in the lung, and spleen. It regulates G protein-coupled receptor signaling cascades and inhibits signal transduction by increasing the GTPase activity of G protein a subunits, thereby driving them into their inactive GDP-bound form. It also serves as a GDP-dissociation inhibitor (GDI) and thereby modulates signal transduction via G protein a subunits. It is reported to display GDI activity on Gi a subunits GNAI1 and GNAI3, but not on GNAI2 and Go a subunit GNAO1. RGS14 also serves as a positive modulator of microtubule polymerization and spindle organization through a Gi a -dependent mechanism. It is reported to associate with the perinuclear sheaths of microtubules surrounding the pronuclei, prior to segregating to the anastral mitotic apparatus. RGS14 contains a RGS domain (aa 67-184) that is essential for its GTPase-activating protein (GAP) activity for G subunits and localization to the nucleus and centrosomes and its GoLoco domain (aa 497-519) is shown to be necessary for its GDI activity and, translocation out of the nucleus and interaction with GNAI1, GNAI2 and GNAI3. It also has two RBD domains (aa 300-371 and 373-443) that are also essential for its localization to the nucleus and centrosomes.
