      Background InformationNCoA2 (nuclear receptor coactivator 2) is a transcriptional coactivator for steroid receptors and nuclear receptors. Also known as glucocorticoid receptor interacting protein 1 (GRIP1), steroid receptor coactivator-2 (SRC-2), or transcriptional mediators/intermediary factor 2 (TIF2), this coregulatory protein contains several nuclear receptor interacting domains and an intrinsic histone acetyltransferase activity. NCoA-2 assists nuclear receptors in the upregulation of DNA expression. In cells ligand activated nuclear receptors recruit NCoA-2 to DNA where it acylates histones to make targeted regions accessible to transcriptional machinery.

      INVOLVEMENT IN DISEASE: Chromosomal aberrations involving NCOA2 may be a cause of acute myeloid leukemias. Inversion inv8(p11;q13) generates the MYST3-NCOA2 oncogene, which consists of the N-terminus part of MYST3/MOZ and the C-terminus part of NCOA2/TIF2. MYST3-NCOA2 binds to CREBBP and disrupts its function in transcription activation.
