SpecificityThis antibody binds strongly to the HLA-B27 antigen. It also binds less strongly to the HLA-B7 antigen. Affinity studies by Scatchard analysis showed that the antibody has a higher affinity for HLA-B27 (9.7x108M-1) than for HLA-B7

      (9.5x107M-1).

      Cell reactivity:

Flow cytometric analysis of lymphocyte staining for HLA-B27 gave the following results:

      Subjects Fluorescence Intensity

      Heterozygous HLA-B27+ Strong

      Homozygous HLA-B7+ Strong

      Heterozygous HLA-B7+ Faint

      Non B27, Non B7 Negative

      Entrez Gene Number
Entrez Gene SummaryHLA-B belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta-2 microglobulin). The heavy chain is anchored in the membrane. Class I molecules play a central role in the immune system by presenting peptides derived from the endoplasmic reticulum lumen. They are expressed in nearly all cells. The heavy chain is approximately 45 kDa and its gene contains 8 exons. Exon 1 encodes the leader peptide, exon 2 and 3 encode the alpha1 and alpha2 domains, which both bind the peptide, exon 4 encodes the alpha3 domain, exon 5 encodes the transmembrane region and exons 6 and 7 encode the cytoplasmic tail. Polymorphisms within exon 2 and exon 3 are responsible for the peptide binding specificity of each class one molecule. Typing for these polymorphisms is routinely done for bone marrow and kidney transplantation. Hundreds of HLA-B alleles have been described.

SIZE: 362 amino acids; 40478 Da

SUBUNIT: Heterodimer of an alpha chain and a beta chain (beta-2- microglobulin). Interacts with human herpesvirus 8 MIR1 protein (By similarity).

SUBCELLULAR LOCATION: Membrane; Single-pass type I membrane protein.

      DOMAIN: SwissProt: P30498

PTM: Polyubiquitinated in a post ER compartment by interaction with human herpesvirus 8 MIR1 protein. This targets the protein for rapid degradation via the ubiquitin system (By similarity).

DISEASE: "SwissProt: P03989 # HLA-B27 is associated with the development of ankylosing spondylitis (AS) [MIM:106300]. AS is a chronic inflammatory rheumatic disease that mainly affects the axial skeleton and is considered the prototype of seronegative spondyloarthropathies (SNSA), which include reactive arthritis (e.g., Reiter's syndrome), psoritic spondylitis, and other spondyloarthropathies (SpA). In the Greek Cypriot population, a restricted number of HLA-B27 subtypes are associated with AS and other B27-related diseases and an elevated frequency of the B*2702 allele in the AS patients is identified. The allele B*2707 seems to have a protective role in this population because it was found only in the healthy controls.| P18464 # HLA-B51 is associated with Behcet disease but it is not certain whether HLA-B51 itself or a closely linked gene is responsible for susceptibility. About 20% of healthy individuals of various ethnic origin carries the HLA-B 51 locus, compared with 50 to 80% of patients. Behcet disease is a systemic disorder of recurrent acute inflammation characterized by 4 major symptoms: oral aphthous ulcers, skin lesions, ocular symptoms and genital ulcerations. Occasionally, inflammation in tissues and organs throughout the body can occur, including the gastrointestinal tract, central nervous system, vascular system, lungs, and kidneys. The etiology of Behcet disease is unclear.| P30464 # Involved in susceptibility to Stevens-Johnson syndrome [MIM:608579]. This is a life-threatening reaction of the skin to particular types of medication. It is characterized by high fever, malaise and blistering exanthema. The more severe form of the disease is referred to as toxic epidermal necrolysis."
