      Applications
ApplicationAnti-Dopamine D1A Receptor Antibody, C-terminus, cytoplasmic is an antibody against Dopamine D1A Receptor for use in ELISA, IH & WB.
      Key Applications
      • ELISA
      • Immunohistochemistry
      • Western Blotting
Application NotesWestern blot: 1-10 μg/mL (Chemiluminescence technique). An antibody made to this peptide has been shown to detect 2 proteins of 50 and 75 kDa in solubilized hippocampal membranes. The 75 kDa band was not visible in striatum (Huang et al. 1992; O'Connel et al. 1995). Approximately 100-400 μg of protein/lane should be loaded to visualize the 75 kDa band (Huang et al. 1992; O'Connel et al. 1995) that is present in relatively small amounts.

Immunohistochemistry: 2-10 μg/mL on paraformaldehyde fixed sections.

      ELISA: 0.5-1.0 μg/mL (1 μg/mL D1A peptide (Cat. Number AG259)/well)

Optimal working dilutions must be determined by end user.
      Biological Information
ImmunogenA 13 amino acid peptide sequence (amino acids 403-415) from rat D1A (Monsma et al. 1990; Zhou et al. 1990; Zhou et al. 1992; O'Dowd et al. 1990). This peptide is predicted to be cytoplasmic.
      EpitopeC-terminus, cytoplasmic
ConcentrationPlease refer to the Certificate of Analysis for the lot-specific concentration.
      HostRabbit
SpecificityDopamine Receptor D1A. An antibody made to the D1R peptide has been shown to detect 2 proteins of 50 and 75 kDa in solubilized hippocampal membranes by Western blot. The 75 kDa band was not visible in striatum (Huang et al. 1992). Approximately 100-400 μg of protein/lane should be loaded to visualize the 75 kDa band. By immunohistochemistry D1R has been localized in various regions of the rat brain (Huang et al. 1992).

May recognize porcine or bovine (100 % similarity) or human (78% similarity).

      Entrez Gene Number
Entrez Gene SummaryThis gene encodes the D1 subtype of the dopamine receptor. The D1 subtype is the most abundant dopamine receptor in the central nervous system. This G-protein coupled receptor stimulates adenylyl cyclase and activates cyclic AMP-dependent protein kinases. D1 receptors regulate neuronal growth and development, mediate some behavioral responses, and modulate dopamine receptor D2-mediated events. Alternate transcription initiation sites result in two transcript variants of this gene.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: P21728 # This is one of the five types (D1 to D5) of receptors for dopamine. The activity of this receptor is mediated by G proteins which activate adenylyl cyclase.
SIZE: 446 amino acids; 49293 Da
SUBUNIT: Interacts with DNAJC14 via its C-terminus (By similarity). Interacts with DRD1IP.
SUBCELLULAR LOCATION: Cell membrane; Multi-pass membrane protein (By similarity). Endoplasmic reticulum membrane; Multi-pass membrane protein (By similarity). Note=Transport from the endoplasmic reticulum to the cell surface is regulated by interaction with DNAJC14 (By similarity).
TISSUE SPECIFICITY: Detected in caudate, nucleus accumbens and in the olfactory tubercle.
SIMILARITY: SwissProt: P21728 ## Belongs to the G-protein coupled receptor 1 family.
