      DescriptionAnti-CLSTN3b
Background InformationCalsyntenin 3beta (UniProt: A0A4P8DYG6; also known as Calsyntenin-3β, CLSTN3β) is encoded by the Clstn3 (also known as Clstn3b) gene (Gene ID: 232270) in murine species. CLSTN3β is a mammal-specific that is preferentially expressed in thermogenic adipocytes and is localized in endoplasmic reticulum. Its expression in brown adipose tissue is about six-fold higher than both the inguinal subcutaneous and the perigonadal visceral white adipose tissue. However, its expression can be induced in the inguinal subcutaneous white adipose tissue upon cold exposure. Brown adipose tissue thermogenesis require innervation by the sympathetic nerves that release norepinephrine, which activates β-adrenergic receptors-cAMP-PKA pathway to drive lipolysis and thermogenic respiration. CLSTN3β is shown to promote S100b protein release that promotes this innervation and its loss is shown to reduce functional sympathetic innervation in adipose tissue. Loss of CLSTN3β also predisposes mice to obesity on a high fat diet. Clstn3β knockout mice exhibit significantly higher fasting glucose levels, impaired glucose tolerance, more lipid accumulation in brown adipose tissue, and liver steatosis. Applications
ApplicationAnti-CLSTN3b, Cat. No. ABS2236, is a rabbit polyclonal antibody that detects Calsyntenin 3b (Clstn3) and is tested for use in Immunofluorescence, Immunoprecipitation, and Western Blotting.
      Key Applications
      • Western Blotting
      • Immunoprecipitation
      • Immunofluorescence
      Application NotesTested Applications

Evaluated by Western Blotting in lysate from wild-type brown adipocytes.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected Calsyntenin 3b (Clstn3) in lysate from wild-type adipocytes, but not in lysate from Calsyntenin 3b knock out adipocytes.

Immunoprecipitation Analysis: A representative lot immunoprecipitated CLSTN3b in Immunoprecipitation applications (Zeng, X., et. al. (2019). Nature. 569(7755): 229-235).

Western Blotting Analysis: A representative lot detected CLSTN3b in Western Blotting applications (Zeng, X., et. al. (2019). Nature. 569(7755): 229-235).

Immunofluorescence Analysis: 10 ug/mL from a representative lot detected CLSTN3b in mouse primary brown adipocytes (Courtesy of Dr. Xing Zeng @UT Southwestern Medical Center).

Immunofluorescence Analysis: A representative lot detected CLSTN3b in Immunofluorescence applications (Zeng, X., et. al. (2019). Nature. 569(7755): 229-235).
