Application NotesWestern blot: 0.1 μg/mL on Triton soluble fraction of rat brain. Suggested dilution buffer is TBS-T and blocking buffer is 3% skim milk. Suggested incubation time is 3 hours to overnight at room temperature.
Immunohistochemistry: 0.25-0.5 μg/mL on 4% paraformaldehyde fixed tissue. Suggested dilution buffer is PBS-T. Suggested permeabilization method is 0.1% Triton X-100. Suggested incubation time is 2 hours to overnight at room temperature.
      Immunocytochemistry
Immunoprecipitation: 1-2 μg. Suggested reaction volume is 200-1000 μL. Suggested total protein concentration is 2 mg/mL. Suggested incubation time is 2 hours at 2-8°C. Suggested capture agent is Protein A.

Optimal working dilutions must be determined by end user.
      Biological Information
ImmunogenGST fusion protein from rat stargazin, cytoplasmic domain.
      HostRabbit
SpecificityStargazin [Stargazin-like TARP (transmembrane AMPA receptor regulatory protein)]. By Western blot the antibody recognizes bands at ~35 kDa and ~55 kDa on Triton soluble fraction of rat brain. The antibody reacts dominantly with TARP gamma-8 and weakly with gamma-3 and gamma-4. Stargazin is expressed strongly in the hippocampus and cerebellum.

      Entrez Gene Number
Entrez Gene SummaryL-type calcium channels are composed of five subunits. The protein encoded by this gene represents one of these subunits, gamma, and is one of several gamma subunit proteins. It is an integral membrane protein that is thought to stabilize the calcium channel in an inactive (closed) state. This protein is similar to the mouse stargazin protein, mutations in which have been associated with absence seizures, also known as petit-mal or spike-wave seizures. This gene is a member of the neuronal calcium channel gamma subunit gene subfamily of the PMP-22/EMP/MP20 family.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: Q9Y698 # Thought to stabilize the calcium channel in an inactivated (closed) state.
SIZE: 323 amino acids; 35966 Da
SUBUNIT: The L-type calcium channel is composed of five subunits: alpha-1, alpha-2/delta, beta and gamma. Interacts with the PDZ domains of DLG4/PSD-95 and DLG1/SAP97. May interact with GOPC (By similarity).
SUBCELLULAR LOCATION: Membrane; Multi-pass membrane protein.
      TISSUE SPECIFICITY: Brain.
PTM: Phosphorylation of Thr-321 impairs interaction with DLG1 and DLG4.
SIMILARITY: SwissProt: Q9Y698 ## Belongs to the PMP-22/EMP/MP20 family. CACNG subfamily.
