Background InformationG9a (BAT8 or Histone-lysine N-methyltransferase) is thought to be involved in intracellular protein-protein interaction and is a lysine-preferring histone methyltransferase. H3 Lys-9 methylation represents a specific tag for epigenetic transcriptional repression by recruiting HP1 proteins to methylated histones. The presence of the ankyrin repeats also suggests that G9a is involved in intracellular protein-protein interaction. Like Suv39h1, G9a transfers methyl groups to the lysine residues of histone H3, but with a 10- to 20-fold higher activity. G9a is localized to the nucleus but not in the repressive chromatin domains of centromeric loci, in which Suv39h1 family proteins were localized. This protein is probably targeted to histone H3 by different DNA-binding proteins like E2F6, MGA, MAX and/or DP1. G9a also methylates histone H1.

      Entrez Gene Number
Entrez Gene SummaryA cluster of genes, BAT1-BAT5, has been localized in the vicinity of the genes for TNF alpha and TNF beta. This gene is found near this cluster; it was mapped near the gene for C2 within a 120-kb region that included a HSP70 gene pair. These genes are all within the human major histocompatibility complex class III region. This gene was thought to be two different genes, NG36 and G9a, adjacent to each other but a recent publication shows that there is only a single gene. The protein encoded by this gene is thought to be involved in intracellular protein-protein interaction. There are three alternatively spliced transcript variants of this gene but only two are fully described.

      UniProt Number
UniProt SummaryFUNCTION: Histone methyltransferase. Preferentially methylates 'Lys-9' of histone H3 and 'Lys-27' of histone H3 (in vitro). H3 'Lys-9' methylation represents a specific tag for epigenetic transcriptional repression by recruiting HP1 proteins to methylated histones. Probably targeted to histone H3 by different DNA-binding proteins like E2F6, MGA, MAX and/or DP1. Also methylates histone H1 By similarity.
      CATALYTIC ACTIVITY: S-adenosyl-L-methionine + histone L-lysine = S-adenosyl-L-homocysteine + histone N(6)-methyl-L-lysine.
      SUBUNIT STRUCTURE: Part of the E2F6.com-1 complex in G0 phase composed of E2F6, MGA, MAX, TFDP1, CBX3, BAT8, EUHMTASE1, RING1, RNF2, MBLR, L3MBTL2 and YAF2. Interacts with GFI1B, WIZ and EHMT1.
SUBCELLULAR LOCATION: Nucleus. Note: Associates with euchromatic regions. Does not associate with heterochromatin.
TISSUE SPECIFICITY: Expressed in all tissues examined, with high levels in fetal liver, thymus, lymph node, spleen and peripheral blood leukocytes and lower level in bone marrow.
DOMAIN: The SET domain mediates interaction with WIZ.
PTM: Phosphorylated upon DNA damage, probably by ATM or ATR.
SEQUENCE SIMILARITIES: Belongs to the histone-lysine methyltransferase family. Suvar3-9 subfamily.
      Contains 7 ANK repeats.
      Contains 1 post-SET domain.
      Contains 1 pre-SET domain.
      Contains 1 SET domain.
CAUTION: reported that while NG36 and G9a were originally thought to derive from two separate genes, all G9A transcripts also contain the in frame coding sequence of NG36.
It is uncertain whether Met-1 or Met-21 is the initiator methionine.
SEQUENCE CAUTION: The sequence AAD21811.1 differs from that shown. Reason: Erroneous gene model prediction.
The sequence AAD21812.1 differs from that shown. Reason: Erroneous gene model prediction.
The sequence BAB63294.1 differs from that shown. Reason: Erroneous gene model prediction.
The sequence BAB63295.1 differs from that shown. Reason: Erroneous gene model prediction.
