      Background InformationB-cell lymphoma/leukemia 10 (Bcl-10) is a protein component of a membrane raft complex located within the cytoplasm. This complex is comprised of several proteins including Bcl-10, IKBKB, DPP4 and CARD11. Bcl-10 contains a single CARD domain that allows it to interact with other CARD-proteins including CARDs 9, 10, 11 and 14. It also forms a strong complex with MALT1. Bcl-10 is a promoter of NF-kappa-b activation through NIK and IKK pathways, pro-caspase-9 activation, and apoptosis. It may also serve a role as an adaptor protein for the upstream complex TNFR1-TRADD-Rip and the downstream complex NIK-IKK-IKAP, as well as being a substrate for MALT1. Expression of Bcl-10 is ubiquitous in tissue.

Although the BCL10/IgH translocation leaves the coding region of BCL10 intact, frequent BCL10 mutations could be attributed to the Ig somatic hypermutation mechanism resulting in nucleotide transitions.

      Defects in BCL10 are involved in various types of cancer.
