      Background InformationZinc finger CCCH-type antiviral protein 1 (UniProt: F7F0B1; also known as ZAP) is encoded by the Zc3hav1 gene (Gene ID: 252832) in rat. ZAP acts as a host antiviral factor that inhibits the replication of several diverse viruses by preventing the accumulation of viral RNA in the cytoplasm. It is a member of the poly(ADP-ribose) polymerase family and is alternatively spliced. Its long isoform (ZAPL) contains a PARP-like domain on the C-terminal region, which is absent in the short isoform (ZAPS). ZAPL is shown to be more active against alphaviruses, such as SINV and Semliki Forest virus, than ZAPS. Although both isoforms are shown to be induced by interferon, ZAPS is up-regulated more than ZAPL by virus infection and type I interferon. Over-expression of ZAP is reported to impart a cellular antiviral state against Retroviridae, Togaviridae, and Filoviridae virus family members and this effect exists even in the absence of interferon treatment of cells. TRIM25 is shown to enhance the antiviral action of ZAP via interaction with SPRY domain. TRIM25 mutants that lack the RING or coiled coil domain fail to stimulate antiviral activity of ZAP. The N-terminal region of ZAP (254 amino acids; NZAP) is also shown to exhibit antiviral activity in rat and human cells. (Ref.: Li, MMH., et al. (2016). PLoS Pathog. 13(1): e1006145).
