      Background InformationTropomyosin alpha-4 chain (UniProt: P09495; also known as Tropomyosin-4, TM-4) is encoded by the Tpm4 gene (Gene ID: 24852) in rat. Tropomyosins are dimers of coiled-coil proteins that provide stability to actin filaments and regulate access of other actin-binding proteins. Tropomyosin-4 1 is a cytoskeletal protein that associates with F-actin stress fibers and binds to actin filaments in muscle and non-muscle cells. It plays a central role, in association with the troponin complex, in the calcium dependent regulation of vertebrate striated muscle contraction. It has a coiled coil structure formed by two polypeptide chains. It is abundantly expressed in kidney, lung, and spleen and is also observed in muscle and stomach tissues. Loss of Tpm4.1 is reported to lead to disturbance in cell-cell adhesions and invasive behavior in breast epithelial cells through Rac1-myosin IIB signaling. In colon cancer cells, TPM4 mRNA and protein expressions is shown to be significantly reduced and its low expression correlates with the clinal stage. Clone 2D2A12 detects both rat and human tropomyosin-4 and does not react with other exon1b containing isoforms from other tropomyosins. (Ref.: Schevzov, G., et al. (2011).
