      Background InformationPotassium channel subfamily K member 4 (UniProt: O88454; also known as TWIK-related arachidonic acid-stimulated potassium channel protein, TRAAK) is encoded by the Kcnk4 (also known as Traak) gene (Gene ID: 16528) in murine species. TRAAK is a multi-pass membrane protein that is expressed in brain, spinal cord, and eye and serves as a voltage-insensitive potassium channel. It is a mechano-activated channel that senses shear stress and negative membrane pressure. It generates progressive and non-inactivating current when negative pressure is applied to the plasma membrane. The channel opening is triggered by mechanical forces that deform the membrane. Channel opening is also triggered by raising the intracellular pH to basic levels. TRAAK channel is shown to be inactive at 24C, but raising the temperature to 37C or higher increases the frequency of channel opening. It plays a role in the sensory perception of pain caused by heat. The C-terminal of TRAAK has a charged cluster that is important in maintaining the mechanosensitive properties of the channel. TRAAK channels are reported to be gradually and reversibly activated by polyunsaturated fatty acids (PUFA) including arachidonic acid that potentiates TRAAK background currents by 5- to 20-fold.
