      Background InformationProtein-arginine deiminase (UniProt: A0A5F9DB16; also known as PADI2, PAD2) is encoded by the PADI2 gene (Gene ID: 100357368) in rabbit. Protein-arginine deiminases (PAD) catalyze the hydrolysis of peptidyl-arginine to peptidyl-citrulline and alter protein function. Five isoforms of PAD have been described in mammals (PAD1, 2, 3, 4, and 6) that are homologous in their catalytic site but are expressed differently in different cells and tissues and have different physiological roles. PAD2, one of the most abundant PADs has three domains: N-terminal domain (aa 1-114), a central domain (aa 116-274), and a C-terminal domain (aa 285-657). High calcium concentration is reported to be a prerequisite for PAD2 activation. A redox potential maintains its catalytic site in a reduced state. Overexpression of PAD2 has been associated with inflammatory diseases, such as rheumatoid arthritis and neurodegenerative diseases including multiple sclerosis. Higher levels of PAD2 are observed in transformed cell model of breast cancer progression with increasing tumor malignancy and in a murine models of xenograft tumors.
