      Background InformationNucleus accumbens-associated protein 1 (UniProt: Q96RE7; also known as NAC-1, BTB/POZ domain-containing protein 14B) is encoded by the NACC1 (also known as BTBD14B, NAC1) gene (Gene ID: 112939) in human. NAC-1 serves as a transcriptional repressor in neuronal cells through the recruitment of HDAC3 and HDAC4. It contains a BTB domain (aa 30-94; Broad Complex, Tramtrack and Bric à brac) at its N-terminal and a BEN domain (aa 374-471; BANP, E5R and NAC1) in its C-terminal region. NAC1 undergoes homodimerization through its BTB domain. It differs from other BTB-domain transcription factors in that it lacks a characteristic zinc finger or basic leucine-zipper DNA-binding domain. NAC-1 is shown to be important in maintaining pluripotency of embryonic stem cells. It can direct bind to Nanog and also regulates the expression of Nanog, Oct4, and Sox-2. NAC-1 is reported to be overexpressed in several types of human carcinomas, including ovarian serous, pancreatic, and breast carcinoma. It contributes to tumor progression, tumor cell proliferation, and survival through repressing transcriptional activity of GADD45GIP1. NAC-1 knock-down can result in the apoptosis of ovarian cancer cell lines and enhance their chemosensitivity. (Ref.: Okazaki, K., et al. (2012).
