Background InformationMouse Interferon regulatory factor 7 or IRF-7 (UniProt P70434) is encoded by the murine irf7 gene (Entrez Gene ID 54123). IFN-γ-induced gene activations are mediated by both signal transducers and activators of transcription (STATs) and interferon regulatory factors (IRFs). STAT1 dimer, for example, stimulates immediate transcription by binding IFN-γ-activated site (GAS) sequence in the promoter region of target genes. While a delayed, secondary transcription activation is observed among IFN-γ target genes containing an IFN regulatory factors (IRF) binding site within the interferon-stimulated response element (ISRE) sequence. Irf7 transcription is shown to be activated by STAT1 and IRF9 in response to type II interferon (IFN) IFN-γ stimulation in MEF cultures. The expressed IRF7 protein then further cooperates with STAT1 and IRF1 to stimulate the expression of a subset of IFN-γ-induced STAT1 target genes (PMID 22252317). Cat. No. ABF130, Anti-IRF7 is a rabbit polyclonal antiserum raised against recombinant GST fusion protein (PMID 22252317) encompassing murine IRF7 C-terminal half sequence. This antiserum specifically detects INF-gamma-induced IRF7 expression and chromatin association in Wt, but not Irf7-/- MEF by Western blotting and ChIP application, respectively (PMID 22252317). Murine IRF7 has a calculated M.W. of 51.2 kDa, but migrates with an apparent M.W. of ~60 kDa by SDS-PAGE.

      Key Applications
      • Western Blotting
      • Chromatin Immunoprecipitation (ChIP)
Application NotesChromatin Immunoprecipitation Analysis: A representative lot immunoprecipitated IRF7-associated DNA fragments using chromatin prepared from infected (12 hours with Vesicular Stomatitis Virus/VSV) or uninfected MEFs cells (Thomas Decker, Max F. Perutz Laboratories)
Western Blotting Analysis: A representative lot detected IRF7 in IFNgamma treated WT MEF (Farlik, M., et al. (2012) Mol. Cell Biol. 32(6):1032-1043).
Chromatin Immunoprecipitation Analysis: A representative lot detected IFNgamma-induced IRF7 chromatin association by ChIP using WT MEFs, Tbk1-/Ikbke-MEFs, and Irf7-/-MEFs (Farlik, M., et al. (2012) Mol. Cell Biol. 32(6):1032-1043).
