Background InformationElongation of very long chain fatty acids protein 3 (UniProt: Q9HB03; also known as EC:2.3.1.199, 3-keto acyl-CoA synthase ELOVL3, Cold-inducible glycoprotein of 30 kDa, ELOVL fatty acid elongase 3, ELOVL FA elongase 3, Very long chain 3-ketoacyl-CoA synthase 3, Very long chain 3-oxoacyl-CoA synthase 3) is encoded by the ELOVL3 (also known as CIG30) gene (Gene ID: 83401) in human. ELOVL3 is a multi-pass membrane protein of endoplasmic reticulum (ER). It catalyzes the first and rate-limiting reaction of the four reactions that constitute the long-chain fatty acids elongation cycle. Its C-terminal di-lysine motif (aa 266-270) confers endoplasmic reticulum localization. This ER-bound enzymatic process allows the addition of 2 carbons to the chain of long- and very long-chain fatty acids (VLCFAs) per cycle. It exhibits higher activity towards C18:0 acyl-CoAs. It is strongly expressed in normal skin, breast, colon, liver tissues. A decrease in the expression of ELOVL3 is reported in colorectal tumors compared to normal colorectal epithelium. ELOVL3 is reported to be involved in the regulation of the progression of adipogenesis through activation of peroxisome proliferator-activated receptor (PPAR) in mouse adipocytic 3T3-L1 cells. Its levels are elevated more than200-fold in cold-stressed animals. (Ref.: Alnabulsi, A., et al. (2019). Int. J. Cancer. 145(4);1138-1147; Kobayashi, T., and Fujimori, K.
