      DescriptionProtein Tyrosine Phosphatases (PTPs) and Protein Tyrosine Kinases (PTKs) are gene families encoding important proteins involved in signal transduction. PTPs can be classified as either tyrosine-specific or dual-specific based on their phosphatase activity. Diversity of PTP family includes both receptor -like transmembrane proteins and soluble cytosolic proteins. Each PTP contains a highly conserved catalytic domain of ~240 residues that share high sequence similarity throughout the family. The structures of PTP catalytic domains are highly conserved. Hence, the structural and functional diversity within the family depend on structures of noncatalytic regulatory and targeting domains attached to the N- and C- terminals of the catalytic domain. The catalytic site of PTP resides within a crevice on the molecule's surface. The base of the catalytic site is formed by the residues His214 through Arg221.

      PTP inhibitors serve as valuable tools in studying the functions of these enzymes as also serve as potential therapeutic agents. To assess the physiological function of PTPases, specific and selective inhibitors of peptidic and nonpeptidic nature have been developed.
