      Background InformationTumor suppressor ARF (UniProt Q8N726; also known as Alternative reading frame, ARF, Cyclin-dependent kinase inhibitor 2A, p14ARF, p19ARF) is encoded by the CDKN2A (also known as CDKN2, MLM) gene (Gene ID 1029) in human. p14ARF acts as a tumor suppressor and a key regulator of cellular proliferation that is frequently inactivated in human cancers. It binds to MDM2 and blocks its oncogenic action by inhibiting MDM2-induced degradation of p53. Reported to form a ternary complex with p120(E4F) and p53 that enhances p14ARF-induced G2 cell cycle arrest in a p53-dependent manner. However, it can also induce G2 arrest and apoptosis in a p-53-independent manner by preventing the activation of cyclin B1/CDC2 complexes. Binds to E2F1 and MYC and blocks their transcriptional activator activity without affecting MYC transcriptional repression. Human p14ARF is shown to promote the proteasomal degradation of Melanoma antigen-A11 (MAGE-A11) independent of HDM2 E3 ubiquitin ligase or lysine ubiquitination. It inhibits the ability of MAGE-A11 to stabilize and stimulate androgen receptor (AR) or E2F1 transcriptional activity. Ref.: Minges, JT et al. (2015). J. Biol. Chem. 290, 25174-25187.       Application NotesImmunohistochemistry Analysis: A representative lot detected p14ARF (p19ARF) immunoreactivity in formalin-fxed, paraffin-embedded human lung tissue sections. An upregulated p14ARF (p19ARF) immunoreactivitiy was found in a lung tissue with squamous cell carcinoma when compared to a normal lung tissue sample (Vonlanthen, S., et al. 1998. Oncogene. 17(21):2779-2785).

      Immunoprecipitation Analysis: A representative lot co-immunoprecipitated MDM2 and p53 with p14ARF (p19ARF) either from HeLa cells transfected to overexpress the three proteins or from in vitro transcription/translation reactions (Zhang, Y., et al. 1998. Cell. 92(6):725-234).
