      Background InformationNitric oxide (NO) is an inorganic, gaseous free radical that carries a variety of messages between cells. Vasorelaxation, neurotransmission and cytotoxicity can all be potentiated through cellular response to NO. NO production is mediated by members of the nitric oxide synthase (NOS) family. NOS catalyzes the oxidization of L-arginine to produce L-citrulline and NO. Two constitutive isoforms, brain or neuronal NOS (b or nNOS, type I) & endothelial cell NOS (eNOS, type III), and one inducible isoform (iNOS, type II), have been cloned. Cytokines such as interferon-gamma (IFN), tumor necrosis factor (TNF), interleukin-1 and -2, and lipopolysaccarides (LPS) cause an increase in iNOS mRNA, protein, and activity levels. Protein kinase C-stimulating agents exhibit the same effect on iNOS activity. Human iNOS is regulated by calcium/calmodulin (in contrast with mouse NOS2).

      Application NotesImmunohistochemistry:
      A previous lot of this antibody has been reported to detect iNOS/NOS II in 2% paraformaldehyde-fixed sheep lung (Sherman, T.S., 1999).

      Immunoprecipitation:
      4 μg of a previous lot immunoprecipitated iNOS/NOS II from 500 μg of L929B cells which had been stimulated with 10 ng/mL IFNg and 1 μg/mL LPS.

      Quality AssuranceRoutinely evaluated by immunoblot on lysates from L929B cells that had been stimulated IFNγ and LPS.

      Western Blot Analysis: 1-4 μg/mL of this lot detected iNOS/NOS II in RIPA lysates from RAW 264.7 cells that had been stimulated with LPS. 2 μg/mL of a previous lot detected iNOS/NOS II from similarly treated L8 cells.
