      Background InformationTropomyosin alpha-3 chain (UniProt: Q63610; also known as Gamma-tropomyosin, Tropomyosin-3, Tropomyosin-5, Tpm3) is encoded by the Tpm3 (also known as Tpm-5, Tpm5) gene (Gene ID: 117557) in rat. Tropomyosins are rod-like dimers of coiled-coil proteins that provide stability to actin filaments and regulate access of other actin-binding proteins. In muscle cells, they regulate muscle contraction by controlling the binding of myosin heads to the actin filament. Tropomyosin-3 is a homodimeric protein in slow-twitch skeletal muscle that can also form heterodimers of an alpha (TPM1, TPM3 or TPM4) and a beta (TPM2) chain. It binds to actin filaments in muscle and non-muscle cells and plays crucial role in the calcium dependent regulation of vertebrate striated muscle contraction. In non-muscle cells is implicated in stabilizing cytoskeleton actin filaments. Mutations in Tpm3 gene are associated with diseases of skeletal muscle such as nemaline myopathy, distal arthrogryposis, and the cap disease that is characterized by myopathy and hypotonia that progresses over time. Clone 4D10A5 recognizes the mammalian tropomyosin exon 9a present in muscle and cytoskeletal isoforms produced by the TPM1, TPM2 and TPM3 genes. (Ref.: Schevzov, G., et al. (2011).
