      BackgroundThe mammalian Toll-like receptor (TLR) family is comprised of a group of transmembrane proteins characterized by multiple copies of leucine-rich repeats in the extracellular domain, and an IL-1 receptor motif in the cytoplasmic domain. TLRs signal through adaptor molecules and may be an important component of innate immunity in humans. The TRL family is a phylogenetically conserved mediator of innate immunity and is essential to microbial recognition. TLRs characterized in the past have been shown to activate the MyD88/interleukin -1 receptor-associated kinase (IRAK) signaling pathway. Ten homologs of TLRs (TLR1-TLR10) have previously been described and the cDNA coding for TLR9 has been identified and isolated. Research has shown that TRL9 acts as a receptor for unmethylated CpG dinucleotides in bacterial DNA. Human and mouse TLR9 share an amino acid identity of 75.5%, and it is highly expressed in the spleen.
