      Background InformationSyndecan-1, also known as CD138, is a transmembrane (Type I) heparan sulfate proteoglycan and is a member of the syndecan proteoglycan family. The syndecans mediate cell binding, cell signaling, and cytoskeletal organization. Bearing properties of both heparan sulfate and chondroitin sulfate, Syndecan-1 functions as a cell surface proteoglycan membrane protein and participates in cell proliferation, cell migration and cell-matrix interactions via its receptor for extracellular matrix proteins. Altered syndecan-1 expression has been detected in several different tumor types.

      Molecular WeightThe expected molecular weight of monomeric Syndecan-1 is ~32 kDa; however, the core protein often migrates as an SDS-stable dimer with an approximate M.W. of 77 kDa. Additionally, a high M.W. form of the protein may often be seen between 500-1000 kDa, likely the mature form of Syndecan-1, with all of its heparan and chondroitin sulfate additions. In whole tissues, this mature form often binds tightly to ECM molecules and may require solublization by urea extraction for additional SDS-PAGE analysis (Wilsie, 2006). Triton X-100 or similar mild detergents are sufficient to release the immature precursor forms of Syndecan-1.
