      DescriptionProtein A purified rabbit polyclonal antibody. Recognizes the ~55 kDa (apparent MW) protein disulfide isomerase protein.
      BackgroundPDI is an abundant component of the ER that catalyzes disulfide bond formation in the biosynthesis of various secretory and cell surface proteins. It is a multi-functional polypeptide identical to the β-subunit of prolyl 4-hydroxylase and a cellular thyroid hormone-binding protein. It has two thioredoxin-like regions, each con-taining a Cys-Gly-His-Cys sequence that may represent two independently acting catalytic sites of PDI activity. The C-terminus of PDI has a KDEL sequence, which acts as retention signal for proteins of the ER lumen. PDI exists as a homodimer of ~107 kDa. Prolyl-4-hydroxylase, an α2β2 tetramer, catalyzes the formation of 4-hydroxyproline in collagen by the hydroxylation of proline residues in peptide linkages. The PDI/β-subunit of prolyl-4-hydroxylase is required for its retention in the ER.
