      Application CommentsA protein database search confirms that this peptide sequence is unique to the P2X4 receptor protein. The signal was completely abolished in immunoblotting by preincubating the purified antibody with blocking peptide at 25 µg/ml. Immunohistochemistry was performed on floating mouse brain sections fixed in 4% paraformaldehyde. Staining was completely abolished by preincubating the affinity purified antibody with control peptide at 10-6M (approximately 2 µg/ml; data not shown). Immunohistochemical staining results correlate well with other independent reports and in situ hybridization studies. Antibody should be titrated for optimal results in individual systems.

      BackgroundExtracellular ATP acts as a signaling molecule to mediate a wide variety of physiological responses through interactions with the P2 family of receptors. P2 receptors have been classified into two groups, the P2X family of ligand-gated ion channels, and the P2Y family of metabotropic G-protein coupled receptors. To date, seven P2X receptor subtypes and eight different P2Y receptor subtypes have been identified. The P2X receptor subunits exhibit approximately 35%-48% identity with each other at the amino acid level, and have been shown to form both homo- and hetereromultimeric complexes. The various P2X receptor subunits exhibit a broad pattern of distribution, being localized to different types of immune cells, glands, smooth muscle, skeletal muscle, and the central and peripheral nervous systems. The P2X4 receptor subunit is predominantly expressed in the central nervous system and has a widespread distribution as determined by in situ hybridization and immunohistochemistry.
