Background InformationDelta-type opioid receptor (UniProt: P32300; also known as D-OR-1, DOR-1, K56, MSL-2) is encoded by the Oprd1 gene (Gene ID: 18386) in murine species. Opioid receptors are seven-transmembrane proteins that which belong to the G protein-coupled receptor (GPCR) superfamily. Three types of opioid receptors have been described and are known as Mu ( ), Kappa ( ), and Delta ( ) receptors. Following binding to exogenous opiates and endogenous opioid peptides, such as β-endorphin, enkephalins and dynorphins, they are activated and transmit signals to multiple intracellular effectors, such as ion channels and second messengers that ultimately reduce neuronal activity. These receptors have a common opioid binding pocket within the helical transmembrane core. Opioid receptor delta (DOP) is a multi-pass transmembrane glycoprotein with four extracellular domains, seven transmembrane domains, and four cytoplasmic domains. They are highly concentrated in the basal ganglia and limbic regions of the brain. DOP are expressed in structures of both the ascending and the descending pain pathways. They participate not only in the control of pain, but are also involved in mood disorders, including anxiety and depression. Selective activation of the DOP is reported to have great potential for the treatment of chronic pain. (Ref.: Quirion, B., et al. (2020). Front. Mol. Neurosci. 13; 52; Lutz, PE., and Kieffer, BI.
