      Background InformationNatural cytotoxicity triggering receptor 2 (UniProt: O95944; also known as Lymphocyte antigen 95 homolog, NK cell-activating receptor, Natural killer cell p44-related protein, NK-p44, NKp44, CD336) is encoded by the NCR2 (also known as LY95) gene (Gene ID: 9436) in human. NKp44 is an activating receptor expressed on human NK cells and its expression is induced upon activation by interleukin-2. Its expression is not observed on the surface of resting NK cells. It can induce cytokine production and cytolytic activity in human NK cells. NKp44 consists of a single extracellular V-type Ig domain, a transmembrane region, and a short cytoplasmic. Through its transmembrane region, it associates with the adaptor molecule DNAX activation protein 12 (DAP12) that contains an immunoreceptor tyrosine-based activation motif (ITAM) and DAP12 is required for NKp44 cell-surface expression and downstream signaling events. NKp44 is reported to mediate cytotoxicity against various tumor cells, including neuroblastoma and choriocarcinoma. In human NK-like cell line NKp44 exhibits strong redirected cytotoxicity and modest IFN-gamma production. (Ref.: Fuchs, A., et al. (2005). Blood. 115(6); 2076-2082; Kikuchi-Maki. A., et al. (2003).
