      Background InformationMajor intrinsically disordered NOTCH2-binding receptor 1-like (UniProt: P59773; also known as Membrane integral NOTCH2-associated receptor 2) is encoded by the MINAR2 (also known as KIAA1024L) gene (Gene ID: 100127206) in human. MINAR2, a highly conserved, intrinsically disordered member of the MINAR family of proteins, has an extracellular domain, a single transmembrane domain, and a few amino acids in its cytoplasmic tail. Its expression is observed in the brain with high levels detected in midbrain, hypothalamus, and the pituitary gland. Loss of MINAR2 is reported to cause motor dysfunctions in murine models that resemble Parkinson's disease. MINAR2 knockout mice display significant alterations in their neuronal function, loss of TH positive neurons in pars compacta, and higher accumulation of -synuclein protein. It is reported to be localized to endoplasmic reticulum (ER) compartments and interacts with NOTCH2. ER stress, NOTCH folding, and its trafficking from the ER have been proposed to play a role in motor deficits associated with progression of Parkinson s disease. MINAR2 has also been linked to progressive hearing loss with alteration sin the stereocilia of hair cells. Loss of MINAR2 is shown to reduce cholesterol localization to the hair bundles in the ear that aggravates hair cell deficits. (Ref.: Bademci, G., et al. (2022) Proc. Natl. Acad. Sci.
