      Background InformationKininogen-1 (UniProt: P01042; also known as Alpha-2-thiol proteinase inhibitor, Fitzgerald factor, High molecular weight kininogen, HMWK, Williams-Fitzgerald-Flaujeac factor, KNG) is encoded by the KNG1 (also known as BDK, KNG) gene (Gene ID: 3827) in human. Kininogen (KNG) is the precursor protein of the kallikrein-kinin system. KNGs are shown to act as inhibitors of proteases. Kininigen-1 is a secreted protein that is synthesized with a signal peptide (aa 1-18), which is subsequently cleaved off to generate the mature form that can be further cleave into Kininigen-1 heavy chain (HMW; aa 19-380), Kininien-1 light chain (LMW; aa 390-644), T-kinin (aa 376-389), Bradykinin (aa 381-389), lysyl-bradykinin (aa 380-389), and Low molecular weight growth-promoting factor (aa 431-434). Kininogen-1 is reported to exert anti-angiogenic properties and block the proliferation of endothelial cells. It has been identified as a serum biomarker for advanced colorectal adenoma and colorectal cancer. The HMW-kininogen plays a role in blood coagulation by helping to position prekallikrein and factor XI next to factor XII. It is shown to inhibit the thrombin- and plasmin-induced aggregation of thrombocytes. Activation of factor XII triggers HMW-kininogen cleavage by plasma kallikrein, which results in the generation of cleaved high molecular-weight kininogen (HKc) and the release of bradykinin. Higher rate of HMW-kininogen and elevated levels of plasma kallikrein activity has been recorded in the plasma of Alzheimer s disease subjects and changes HMW and cleaved kininogen levels have been significantly correlated with dementia and neuritic plaque scores.
