      Background InformationHemagglutinin (UniProt: Q5QQ29; also known as HA) is encoded by the HA gene (Gene ID: 3654260) in Influenza A virus H5N1. Hemagglutinin (HA) is a single-pass type I membrane protein that binds to sialic acid-containing receptors on the cell surface, bringing about the attachment of the virus particle to the cell. This attachment induces virion internalization either through clathrin-dependent endocytosis or through clathrin- and caveolin-independent pathway. It plays a major role in the determination of host range restriction and virulence. HA is shown to be a homotrimer, with each subunit made up of two disulfide-linked polypeptides, HA1 and HA2 and each subunit consists of a membrane-proximal helix-rich stem structure and a membrane-distal receptor binding globular domain. HA is reported to mediate viral entry into host cell in two steps. In the first step, it binds to the host receptor and the virus is endocytosed into the cell. In the second step fusion of the endosomal membrane with the viral membrane occurs to release the viral RNA into the host cell. Clone 9F4 is shown to bind both the native and denatured forms of HA. This antibody does not block the interaction between HA and its receptor, but it prevents the pH-mediated conformational change of HA. It is shown to have a protective prophylactic as well as therapeutic effect viral challenge of mice. Amino acid region 260-269 of HA appear to be essential for binding of this monoclonal antibody.
