      Background InformationDimethylaniline monooxygenase [N-oxide-forming] 5 (EC 1.14.13.8; UniProt P49326; also known as Dimethylaniline oxidase 5, FMO 5, Hepatic flavin-containing monooxygenase 5) is encoded by the FMO5 gene (Gene ID 2330) in human. FMO5 belongs to the family of Flavin-containing monooxygenase (FMO) genes that encode ER membrane flavin-containing monooxygenases. Humans possess five functional FMO genes (FMO1 to FMO5). FMO 1, 2 and 3 catalyze NADPH-dependent oxygenation of a wide array of foreign chemicals, while little is known about FMO 4 expression, substrates or activity. FMO 5 is highly expressed in the liver in human and murine species. FMO 5 is known to catalyze the N-oxygenation of short-chain aliphatic primary amines (e.g., N-octylamine) and the S-oxygenation of S-methyl-esonarimod, an active metabolite of the anti-rheumatic esonarimod. FMO 5 is also demonstrated to catalyze a Baeyer-Villiger oxidation reaction. However, FMO 5 exhibits little or no activity towards classic FMO substrates, such as methimazole, trimethylamine, and benzydamine. Fmo5 gene disruption in mice is reported to result in slowed metabolic aging via pleiotropic effects.
