      Application NotesWestern blot: 1 μg/ml for 2 hours at RT.

      Immunoprecipitation: 2 μg/mg of protein lysate Immunohistochemistry (frozen and paraffin): 2-4 μg/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10 mM citrate buffer, pH 6.0, for 10-20 min. followed by cooling for 20 min. at RT.

      Optimal working dilutions must be determined by end user.

      SpecificityRecognizes a protein of 32 kDa, identified as the Caspase-3 protein. Highly specific to Caspase-3 and shows no cross-reaction with other caspase family members. Caspase-3 is ubiquitously expressed and like other caspases is synthesized as an inactive proenzyme. Upon activation, caspase-3 is cleaved generating two subunits of 17 kDa and 12 kDa respectively. Caspase-3 has been implicated in the induction of apoptosis. Activation of caspase-3 occurs in response to a variety of apoptotic inducers, including Fas-mediated apoptosis.

      Entrez Gene SummaryThis gene encodes a protein which is a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme. This protein cleaves and activates caspases 6, 7 and 9, and the protein itself is processed by caspases 8, 9 and 10. It is the predominant caspase involved in the cleavage of amyloid-beta 4A precursor protein, which is associated with neuronal death in Alzheimer's disease. Alternative splicing of this gene results in two transcript variants that encode the same protein.
