Background InformationCREB-regulated transcription coactivator 1 (CRTC1) or in older literature, Mucoepidermoid carcinoma translocated protein 1 or Transducer of regulated cAMP response element-binding element 1 or Transducer of regulated CREB activity 1 (TORC1), is encoded by the human gene named CRTC1/KIAA0616/MECT1, TORC1, or WAMTP1, and CRTC1 is a transcriptional coactivator for CREB1 protein which activates transcription through binding cAMP response element (CRE) sites on DNA. CRTC1 appears to interact with many signaling and metabolic pathways through its regulation of CREB and thus, CRTC1 is involved in diverse physiological processes including energy homeostasis, cancer and endoplasmic reticulum stress. CRTC1 is a powerful activator of PGC1alpha expression and critical for mitochondrial biogenesis in muscle cells. CRTC1 function also varies by cell type. For example, in muscle cells CRTC1 is critical for mitochondrial development, in hippocampal neurons CRTC1 is important for the maintenance of L-LTP (late phase long-term potentiation), and in cortical neurons CRTC1 is important for dendritic spine growth and early neuronal development. CRTC1 is nuclear localized when dephosphorylated and active. CRTC1 is widely expressed in tissues but highest in developing fetal and adult brain, specifically prefrontal cortex and cerebellum. Interestingly recent research on CRTC1 in worms suggests that inhibiting CRTC1 either through phosphorylation or translation extends lifespan significantly, and thus CRTC1 may be important in combating aging and its effects as well. EMD-Millipore Anti-CRTC1 mouse monoclonal antibody has been tested in western blot against recombinant CRTC1 protein and in fluorescent immunocytochemistry on U251 and NTERA2 cell in culture. In addition the antibody has been successfully tested in flow cytometry on K562 cells in culture.
