      Background InformationBrain-specific angiogenesis inhibitor 1-associated protein 2 (UniProt: Q9UQB8; also known as BAI-associated protein 2, BAI1-associated protein 2, Protein BAP2, Fas ligand-associated factor 3, FLAF3, Insulin receptor substrate p53/p58, IRS-58, IRSp53/58, Insulin receptor substrate protein of 53 kDa, Insulin receptor substrate p53, IRSp53,) is encoded by the BAIAP2 gene (Gene ID: 10458) in human. IRSp53 is a homodimeric protein that serves as a link between the links membrane-bound small G-proteins to cytoplasmic effector proteins. It is shown to be essential for CDC42-mediated reorganization of the actin cytoskeleton and for RAC1-mediated membrane ruffling. It slows down actin filament barbed end growth and this inhibition is relieved by CDC42 and counteracted by VASP, which also binds to IRSp53. It is also reported to play a role in neurite growth. Incorporation of IRSp53 into the post-synaptic density (PSD) enables it to limit the number of post-synaptic glutamate receptors and thereby affect synaptic plasticity. It is detected throughout the cytoplasm in the absence of specific binding partners. Six isoforms of IRSp53 have been described that are produced by alternative splicing. Isoforms 1 and 4 are expressed exclusively in the brain. IRSp53 contains an IMD domain (aa 1-250) and a SH3 domain (aa 374-437). The IMD domain forms a coiled coil and can induce actin bundling and filopodia formation. In the absence of G-proteins, intramolecular interactions between the IMD and the SH3 domains gives rise to an auto-inhibited state of the protein. Interaction of the IMD with RAC1 or CDC42 leads to activation. (Ref.: Sawallisch, C., et al. (2009). J. Biol. Chem. 284(14); 9225-9236).
