      Application CommentsThe epitope is unique for AQP1 and is not present in other known related proteins. It is identical in human, rodent, and cattle, and only slightly different in frog. Recognition of AQP1 can be fully inhibited with the soluble AQP1243-261 blocking peptide. For use as a negative control, preincubate 1 µg blocking peptide with 1 µg antibody for 1 h at room temperature. This antibody is also reported to work with frozen sections. Antibody should be titrated for optimal results in individual systems.

      Special InstructionsDissolve the antibody in 50 µl deionized water to make a 1 mg/ml stock solution. Further dilutions should be made in a physiological buffer containing a carrier protein such as 1-3% BSA. Avoid freeze/thaw cycles of stock solutions. To reduce nonspecific staining, centrifuge all antibody preparations before use (10000 x g for 5 min) to remove any aggregates. Dissolve the peptide in 100 µl deionized water and store in aliquots at -20°C.

      BackgroundAQP1 is a 269 amino acid transmembrane protein belonging to the aquaporin family. AQP1 forms the major water transports pathway of red cells and renal proximal tubules and thin descending limb of Henle, but not collecting duct.
