      Background InformationGamma-secretase subunit APH-1A (UniProt Q96BI3; also known as Anterior pharynx defective 1 homolog A, APH-1a, Aph-1alpha, Presenilin-stabilization factor) is encoded by the APH1A (also known as PSF, CGI-78) gene (ORF UNQ579/PRO1141; Gene ID 51107) in human. Gamma-secretase catalyzes the final cleavage of beta-amyloid precursor protein to generate amyloid-beta peptide, the principal component of amyloid plaques found in Alzheimer's diseased (AD) brains. Gamma-secretase is a complex composed of four integral membrane proteins, presenilin (PS), nicastrin (NCT), PEN-2, and APH-1. The PS component could be either a PS1 or PS2 and the APH component could be either an APH-1a or APH-1b, alternative splicings of these four proteins contribute to further heterogeneity among gamma-secretase complexes. The gamma-secretase complex is assembled in the ER, where NCT and APH-1 form an initial scaffold for the PS holoprotein, which becomes stabilized upon interaction with the NCT/APH-1. Subsequent addition of PEN-2 to this trimeric assembly intermediate triggers the endoproteolytic cleavage of the PS holoprotein. The fourth transmembrane domain (TMD4) of APH-1, the PEN-2 C-terminal end, the NCT TMD domain, and the PS1 C-terminal end are shown to be essential for functional gamma-secretase complex assembly. APH-1a is a seven-transmembrane (a.a. 3-23, 32-52, 69-89, 119-139, 159-179, 187-207, 214-234) protein with a lumenal N-terminus and a cytoplasmic C-terminal tail (a.a. 235-265).
