      DescriptionKlenow Enzyme is the large fragment of DNA polymerase I which can be produced by cleavage of intact DNA Polymerase I with subtilisin. The Klenow fragment exhibits 5’ → 3’ DNA polymerase and 3' → 5' exonuclease activity, but lacks the 5’ → 3’ exonuclease activity of intact DNA polymerase I. Klenow is used to fill in recessed 3’ termini as in end-labeling or in the synthesis of the second strand of cDNA from a single-stranded template. Klenow is also used in dideoxy sequencing reactions.
