      Background InformationPyruvate kinase PKLR (UniProt: P12928-2; also known as EC:2.7.1.40, L-PK, Pyruvate kinase isozymes L/R) is encoded by the Pklr gene (Gene ID: 24651) in rat. Pyruvate kinas (PK) is a key glycolytic enzyme that catalyzes the hydrolysis of phosphoenolpyruvate (PEP) to pyruvate, generating ATP in this process. Four tissue specific isozymes of PK have been described in mammals: L (liver), R (erythrocytes), M1 (muscle) and ubiquitous M2. Except for M1 isozyme all other isozymes display positive cooperative kinetic behavior upon binding the allosteric activator fructose-1,6-diphosphate and in the presence of increasing concentrations of PEP. Phosphorylation of liver PK at serine 12 by protein kinase A (PKA), in response to glucagon, is shown to inhibit its enzymatic activity. Phosphorylation at serine 12 increases the Km for PEP by disrupting an activating interaction between its N-terminal unstructured domain and the main body of the protein. Studies have shown a significant decline in liver pyruvate kinase activity in insulin resistance. Also, it has been reported that phospho-Ser 12 form is significantly reduced in high fat fed mice. However, rats fed a high fat diet, but treated with a pyruvate kinase activator, do not exhibit any significant difference in their phosphor-Ser 12 form. (Ref.: Abulizi, A., et al. (2020).
