Axin-1 (UniProt P57094; also known as Axis inhibition protein 1, Protein Fused) is encoded by the Axin1 (also known as Axin, Fu) gene (Gene ID 12005) in murine species. The expansion of the mammalian cerebral cortex is controlled by a balance between the proliferation and differentiation of intermediate progenitors (IPs). Axin-1 is a Wnt signaling pathway scaffold protein that plays a critical role in orchestrating the signals controlling IP population and the number of differentiated neurons during neurogenesis in the developing cerebral cortex. Axin-1 is phosphorylated at Thr485 by Cdk5, the Thr485 phosphorylation results in Axin-1 translocation from the cytoplasm to nucleus upon neuronal progenitor cell (NPC) differentiation. The interaction between cytoplasmic Axin-1 and GSK-3β is critical for the amplification of the undifferentiated IP pool, whereas the interaction between phosphorylated Axin-1 and β-catenin in the nucleus promotes neuronal differentiation. Cdk5-mediated Axin-1 phosphorylation therefore serves as a molecular switch from amplification to differentiation in NPCs.

Western Blotting Analysis: An 1:1,000 from a representative lot detected a develpmental age-dependent axin-1 phosphorylation level in total brain lysates from E18 mouse embryo, postnatal P10 and adult mice (Courtesy of Professor Nancy Y. This polyclonal antibody detected CDK5/p25-dependent axin-1 Thr485 phosphorylation both in vitro and in vivo (Fang, W.Q., et al. (2011). J. Neurosci. 31(38):13613–13624). The Thr485 designation was originally assigned based on mouse (NP_001153070.1) and rat (NP_077381.1) sequences deposited in NCBI protein database. The numbering can vary depending on species and the source of published sequence. For example, the target resiude corresponds to Thr481 of the human sequence and Thr480 of the mouse and rat sequences reported by UniProt (O15169, O35625 and O70239, respectively).
