      Background InformationUbiquitin conjugation factor E4 B (UniProt O95155; also known as Homologous to yeast UFD2, Homozygously deleted in neuroblastoma 1, Ubiquitin fusion degradation protein 2) is encoded by the UBE4B (also known as E4, HDNB1, UBOX3, UFD2, UFD2A) gene (Gene ID 10277) in human. The turnover of plasma membrane proteins involves internalization and sorting by the endocytic pathway. Ubiquitin modification allows proteins to be recognized by endosomally associated protein complexes, enabling sorting of proteins to be degraded from those to be recycled. The ubiquitin ligase UBE4B is reported to interact with "hepatocyte growth factor-regulated tyrosine kinase substrate" (Hrs) of the "endosomal sorting complex required for transport 0" (ESCRT-0) complex at the endosomal membrane, where UBE4B ubiquitinates internalized EGFR to allow its recognition by ESCRT-0. The deubiquinating enzyme USP8 then associates with ESCRT-0 via binding to "signal-transducing adaptor molecule" (STAM), while UBE4B dissociates from ESCRT-0. ESCRT-I and II then bind to cargo and facilitate membrane budding inward toward the lumen, whereas ESCRT-III mediates the dissociation of ESCRTs from endosomal membranes and membrane scission.
