      Background InformationIntegrin alpha-IIb (UniProt: P08514; also known as GPalpha IIb, GPIIb, Platelet membrane glycoprotein IIb, CD41) is encoded by the ITGA2B (also known as GP2B, ITGAB) gene (Gene ID: 3674) in human. GPIIb is a single-pass type I membrane protein of the integrin alpha chain family. It serves as a receptor of fibronectin, fibrinogen, plasminogen, prothrombin, thrombospondin and vitronectin. It recognizes the sequence R-G-D in a wide array of ligands. . Following activation it brings about platelet/platelet interaction through binding of soluble fibrinogen. This step leads to rapid platelet aggregation, which physically plugs ruptured endothelial cell surface. GPIIb is synthesized with a signal peptide of 31 amino acids that is cleaved to produce the mature form that has an extracellular domain (aa 32-993), a helical domain (aa 994-1019), and a cytoplasmic tail (aa 1020-1039). Three isoforms of GPII b are reported that are generated by alternative splicing. Isoform 1 and 2 have been identified in platelets and megakaryocytes, but not in reticulocytes or in Jurkat and U-937 white blood cell line. Isoform 3 is expressed by leukemia, prostate adenocarcinoma and melanoma cells but not by platelets or normal prostate or breast epithelial cells. Clone PAC- 1 is a pentameric IgM that is reported to bind to the activated platelets, but not to unstimulated platelets.
