      BackgroundNod2 (nucleotide-binding oligomerization domain containing 2; accession no. NP 071445) is a 150 kDa protein that is a member of the Nod-like receptor (NLR) family. It is strongly associated with host responses to intracellular invasion by bacteria and inflammatory responses. Mutations in Nod2 have been associated with the inflammatory disorders, Crohn's disease and Blau syndrome. Nod2 contains two caspase recruitment (CARD) domains at the N-terminus, a NACHT domain in the central region, and six leucine-rich repeats (LRRs) at the C-terminus. Bacterial pathogens are recognized by the host cells extracellularly through Toll-like receptors or intracellularly by a family of cytosolic Nod-like receptors (NLRs). NLRs recognize the microbial molecules that are introduced by the pathogens and/or endogenous factors released from damaged tissue. Nod2 detects intracellular presence of muramyl dipeptide, the largest molecular motif common to Gram-negative and Gram-positive bacteria. After detection of the bacterial product, Nod2 forms a complex with receptor-interacting protein 2 (RIP2) through a CARD-CARD interaction. This complex recruits NEMO (IKK-γ) and mediates the ubiquitin-proteasome-dependent degradation of IκBα, which results in the activation and nuclear traslocation of NF-κB and the upregulation of inflammatory mediators.
