      Key Applications
      • ELISA
      • Western Blotting
Application NotesWestern blot: 1-5 μg/mL using ECL.
Immunohistochemistry: Not tested. Recommended antibody testing dilution: 5-10 μg/mL on paraformaldehyde fixed tissue.
ELISA: 1:10,000-1:100,000 using 50-100 ng control peptide (Catalog number AG219) per well.
Optimal working dilutions must be determined by the end user.
      Biological Information
ImmunogenA 19 amino acid peptide sequence within the cytoplasmic domain #4 of human MCT4 (Wilson et al., 1998; Price et al., 1998).
ConcentrationPlease refer to the Certificate of Analysis for the lot-specific concentration.
      HostRabbit
SpecificityRecognizes human Monocarboxylate Transporter 4 (MCT4). The immunogen shows no significant sequence homology with other MCT. MCT4 is prominently expressed in skeletal muscle and other cells with high glycolytic rate such as tumors cells and white blood cells suggesting an important role in lactic acid efflux.

Note: According to new nomenclature, the old MCT3 has now been reclassified as MCT4. Consult Halestrap and Price (1999) Biochem. J. 343:281-299.

      Entrez Gene Number
Entrez Gene SummaryLactic acid and pyruvate transport across plasma membranes is catalyzed by members of the proton-linked monocarboxylate transporter (MCT) family, which has been designated solute carrier family-16. Each MCT appears to have slightly different substrate and inhibitor specificities and transport kinetics, which are related to the metabolic requirements of the tissues in which it is found. The MCTs, which include MCT1 (SLC16A1; MIM 600682) and MCT2 (SLC16A7; MIM 603654), are characterized by 12 predicted transmembrane domains (Price et al., 1998 [PubMed 9425115]).[supplied by OMIM]

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: O15427 # Proton-linked monocarboxylate transporter. Catalyzes the rapid transport across the plasma membrane of many monocarboxylates such as lactate, pyruvate, branched-chain oxo acids derived from leucine, valine and isoleucine, and the ketone bodies acetoacetate, beta-hydroxybutyrate and acetate (By similarity).
SIZE: 465 amino acids; 49469 Da
SUBCELLULAR LOCATION: Cell membrane; Multi-pass membrane protein.
TISSUE SPECIFICITY: Highly expressed in skeletal muscle.
SIMILARITY: SwissProt: O15427 ## Belongs to the major facilitator superfamily. Monocarboxylate porter (TC 2.A.1.13) family.
