ID4 is an inhibitor of differentiation protein, which, along with ID1, ID2 and ID3, plays a key role in the regulation of lineage commitment, cell fate decisions and in the timing of differentiation during neurogenesis, lymphopoiesis and neovascularisation (angiogenesis). ID proteins, induced by the bone morphogenic protein-SMAD pathway, have also been shown to cooperate with LIF to ensure that embryonic stem cells opt for self-renewal.

Transcription factors containing a basic helix-loop-helix (bHLH) motif regulate expression of tissue-specific genes in a number of mammalian and insect systems. DNA-binding activity of the bHLH proteins is dependent on formation of homo- and/or heterodimers. Dominant-negative HLH proteins encoded by Id-related genes, such as ID4, also contain the HLH-dimerization domain but lack the DNA-binding basic domain. Consequently, Id proteins inhibit binding to DNA and transcriptional transactivation by heterodimerization with bHLH proteins (Pagliuca et al., 1995 [PubMed 7665172]).[supplied by OMIM].
FUNCTION: ID (inhibitor of DNA binding) HLH proteins lack a basic DNA-binding domain but are able to form heterodimers with other HLH proteins, thereby inhibiting DNA binding.
SUBUNIT STRUCTURE: Heterodimer with other HLH proteins.
Evaluated by Western Blot in human lung tissue lysate.
Western Blot Analysis: 1 µg/mL of this antibody detected ID4 on 10 µg of human lung tissue lysate.
